From your question itself I can tell you that you aren't a avid football fan (P.S sorry to say this no offense but you know it's true). As a true football  fan would never ask this question as he knows that players like Messi (One Club Players) will never transfer to other clubs in the same league or country in which their home club plays and definitely (as you ask in the question)not to direct rivals .Sometimes they are too loyal to  even tranfer to other clubs in the same continent as they have fear (infact out of respect of their home club) of facing their home club in champions league. Some examples of players loyalty towards their clubs.
Casillas: Tranferred to Porto.

"Wherever I'll go, i will shout: Hala Madrid!"
- Iker Casillas

Xavi : To Al-Sadd
Xavi: "I told Barça I didn't want to join a European club. I don't want to face Barça, I don't want play at Camp Nou with another shirt."

Gerrard: LA Galaxy :needless to say...

Lionel Messi definitely comes under these category of players without a doubt (P.S: I'm a Madridista. Although I have a huge respect for Messi and Iniesta). Messi was, is and will be a Barcelona player until he retires from football (or atleast thats what I hope). Even if he changes the club it will definitely not be in the same league in which  Barcelona plays and it's definitely not going to be Real Madrid.

EDIT: This isn't the entire list of players to explain loyalty. These are just some examples. There are number of other players who remained loyal to their club even after retirement . Some players even terminated their contract so that they won't play for other clubs. Only thing is they aren't quite popular(which they completely  deserve) as these. These days football doesn't quite work like that. A clear vivid example is the departure of San Iker.

As of January 2015, Lionel Messi's transfer value is $262 million (€220 million) (€87 million over Cristiano Ronaldo) and it would just give the rights to the player. There is also a factor of Messi's salary which is currently above $50 million per year. Messi's buy-out clause from Barcelona stands at a whopping €250 million. Even if FFP rules are relaxed, that is still a huge amount for any club.

And there is also this catch:

Barcelona is my life. They have brought me to where I am today. I could not leave, I don’t want to leave. I know the Premier League is very good. But I cannot see myself playing in England because my heart is with Barcelona, always. - Messi

That's a very interesting question, let's analyze various aspects of it:

Typical Transfer: For buying a player as big as Messi, a lot of money is required. By a lot, I mean at least 150million £. That too if and only if Barcelona are willing to sell him, which is next to impossible. Real Madrid can afford that much money but Barcelona will never let that happen.

Activating Buy-out clause: This is a very expensive but effective way of getting a player. Real Madrid used this to get Luis Figo from Barcelona which set the then world record transfer fee. Leonel Messi's buy out clause stands at 250million £ which is very high even for the richest club in the world. This could happen at the cost of selling a few big players and not buying anyone else for the season, That would not be a wise move. So this situation is unlikely to happen.

Messi's desire: In a hypothetical situation where Real can activate buy out clause, I doubt messi would like to move to Santiago Bernabeu. For any player to move to a new club, his desire to play there and achieve bigger things is the most important part of that deal. If the player's heart isn't in the move, then it is a mistake as that player wouldn't play at his best. So, even if somehow Real sign him he won't be as effective as he is in Barcelona and we will get devoid of his magical game. Here, I am logically assuming that Messi doesn't want to play for Real.
